Expression of 2,4 - Dichlorophenoxyacetic Acid Monooxygenase Gene in Tobacco Plants

Abstract
The 2,4-D monooxygenase gene (tfdA) from Alcalegenes eutrophus was modified by PCR primer mutagenesis to produce a plant consensus translational start sequence by changing GTG to ATG. We confirmed the modified gene, mtfdA, by sequencing. The mtfdA gene was cloned into a cauliflower mosaic virus 35S promoter expression vector (pMBP-1/mtfdA) and introduced into tobacco plants by Agrobacterium-mediated transformation. Introduction of the mtfdA gene into genomic DNA of plants was confirmed by Southern blot hybridization analysis. Northern blot hybridization analysis showed that the mtfdA gene was stably expressed in transformed tobacco plants. Transformed tobacco plants tolerated elevated level of 8 §·¡¤L^(-1) 2,4-D in tissue culture and showed resistance when sprayed with 3 g¡¤L^(-1) 2,4-D.
